  1. Flavopiridol induced by adriamycin-resistant Ewing sarcoma cell lines VH-64/ADR apoptosis and mechanism of action

    Investigate the cell cycle inhibitor flavopiridol on adriamycin-resistant Ewing's sarcoma cell proliferation inhibition and apoptosis induction mechanism. Methods: MTT assay adriamycin (ADM) and falvopiridol cells on VH-64/ADR half inhibitory concentration (IC50), calculate the resistance factor; flow cytometry (FCM) detection falvopiridol after administration VH-64 / ADR cell cycle; West-ern-blotting was used to detect cells Bcl-2, pro-caspase-3, activated poly ADP-ribose polymerase (PARP-85) protein. Results: flavopiridol inhibited adriamycin-resistant Ewing's sarcoma cell lines VH / ADR cell proliferation, its effect was time-and concentration correlation (P0.05). flavopiridol administration, the resistant cell line VH-64/ADR apoptosis was significantly higher (P0.05); bcl-2, pro-casepase-3 downregulation and upregulation of PARP activity type. Conclusion: FP VH-64/ADR can effectively induce apoptosis, which may be associated with mitochondrial signal transduction pathway.
